The locking nut is the one in the center.
Try a couple of drops of penetrating fluid.
Is it misthreaded?
Are you turning it in the correct direction?

It has been awhile since I installed the spacer. I cannot remember which direction I turned the nut.

not to hijack this guys thread but i just installed my 5/16 spacer and it may be alla in my head but I feeel a slight vibration in the steering wheel when i get it above like 75. Was this there before and I'm just noticing it now because I'm paranoid? Or do i need to adjust the strut bar?

I just used a regular crescent wrench. Remember to turn it to the LEFT. It's probably pretty tight, but use some force, it should come loose.

if you are turning towards the front of the car, then you would be tightening it against the strut bar, you want to turn it towards the windshield.

Just walked out to the car ---Right hand nut is LEFT HAND THREAD---Left Nut is RIGHT HAND THREAD------U TURN both, left, and right nut ---top towards the front of the car.
In fact if u look at Davidv's pic ---u will see that the right threads are left hand and the left threads are right hand ---On his pic ---turn BOTH towards the lower right hand corner of the pic
