If I have an existing sub installed and replaced it with another sub of a different manufacturer (same size). Do I have to tune the amp to that sub? Or do I just place the new sub in the enclosure and forget about it? Both subs are 300w RMS

than yes, just switch subs if the power ratings are roughly the same.
