Just drive it 500 miles in moderate stop and go traffic.

i replaced both during my install anyways. I drove 1k miles in stop and go traffic just to be on the safe side. Long trips don't count.

I'm doing it at same time that I'm doing the vortech S/C. I have the clutch and flywheel now, just waiting on the S/C. Not replacing tranny, but they are yes on clutch fluid.

I think you misunderstood my question. I will have the clutch, tranny and diff fluid replaced when i have this stuff done. after a 500-1000 mile breakin would i need to replace the fluid again?

Take a look at the clutch fluid after the break-in. If its brownish, replace it otherwise you should be fine. Put in some Valvoline Synpower brake fluid. The tranny and diff fluid should not need to be changed.
