Not necessarily. If you increase the fuel pressure, your stock ECU is going to trim back IPW to get to 14.7:1 at idle and cruise. So the net effect, should be an accurate MPG reading.
But your MPG will still be inaccurate, if using larging injectors and scaling them via MAF adjustment or reflash.
