How long are you visiting for ?
When are you planning to visit the Big Apple ?
What are your interests ?
Where are you specificly staying ?
Not knowing what you are looking to do I'd say Times Square and then branch out from there. Rudy got it all cleaned up years ago.
If you are staying in NYC ( Manhattan ) like was said before, forget the rental, go with taxis, buses and the subway will getcha around, but this time of year the subways are freakin hot hot man. Oh yeah, there is always walking which will give you the flavor of New York, long as it's not too far. You might want to stay above ground getting around.

you NY guys should come out to the meet in Westbury tonight.. http://www.tsz33.com/topic.asp?TOPIC_ID=310&whichpage=1
I'd stay in midtown. Lots of stuff to do there. Times Square, Empires State, and you can just take a cab to downtown and back. Screw the subway...you'll sweat your nuts off this time of year. Midtown is more touristy, while downtown is more beatnik.
You can forget about Niagra that is like forever away from the city.
